that would be done in photoshop using layers and saving it as a .PNG file im assuming.

When using a art program, make the background transparent, then put a picture of whatever on it, when you save it, save it as a PNG.
JPEGs just show transparent parts as white.

GIF files d the same thing, but I see no reason to use it unless there is actually animation in it.
And most people I see do not have animations in pop out siggies.
But you can if you want.
Here is a extremely quick animation I did with it.

Turning things into animations can sometimes lower the quality. o.O
